目的:探讨莪术油对阿霉素诱导耐药的人甲状腺未分化癌细胞株HTh74Rdox增殖和耐药性的影响及机制。方法:用不同浓度莪术油处理HTh74Rdox后观察细胞形态;MTT检测莪术油对HTh74Rdox细胞的生长抑制作用;流式细胞术检测HTh74Rdox细胞凋亡;Western Blot检测Bcl-2和Bax蛋白的表达;q PCR检测MDR1和ABCG2 mRNA的变化。结果:莪术油对HTh74Rdox细胞的增殖有明显抑制作用,联合阿霉素后可减少阿霉素剂量;莪术油明显诱导细胞凋亡(P<0.01);且能够提高Bax/Bcl-2比值(P<0.05),降低MDR1、ABCG2 mRNA表达。结论:莪术油可以抑制HTh74Rdox细胞的生长并改善其耐药作用,其机制可能分别与提高Bax/Bcl-2比值和降低MDR1、ABCG2 mRNA表达有关。
Objective: To investigate the effects and mechanisms of zedoary turmeric oil on the proliferation and drug resistance of adriamycin-resistant human thyroid carcinoma cell line HTh74Rdox. Methods: HTh74Rdox cells were treated with different concentrations of Zedoary turcicum oil. The growth of HTh74Rdox cells was detected by MTT assay. The apoptosis of HTh74Rdox cells was detected by flow cytometry. The expression of Bcl-2 and Bax protein was detected by Western Blot. Detection of MDR1 and ABCG2 mRNA changes. Results: Curcuma oil significantly inhibited the proliferation of HTh74Rdox cells, combined with doxorubicin can reduce doxorubicin dose; Curcuma oil significantly induce apoptosis (P <0.01), and can increase the Bax / Bcl-2 ratio (P <0.05), reduce MDR1, ABCG2 mRNA expression. Conclusion: Curcuma oil can inhibit the growth and improve the drug resistance of HTh74Rdox cells. The mechanism may be related to increasing Bax / Bcl-2 ratio and decreasing the expression of MDR1 and ABCG2 mRNA respectively.
