目的探讨八正散对腺性膀胱炎病理模型大鼠相关细胞因子的影响机制。方法以腺性膀胱炎模型大鼠为研究对象,随机分为空白组、模型组、对照组和八正散的不同剂量(高、中、低)组。对照组给予羟基喜树碱每天3mg/kg膀胱灌注,八正散高、中、低剂量组分别给予同体积八正散液(生药浓度分别为0.84g/mL、0.42g/mL、0.2lg/mL),每日2次,灌注/灌胃连续2周。HE染色观察膀胱病理变化,免疫组织化学法观察膀胱组织中的白细胞介素6(interleukin6,IL-6)、白细胞介素17(interleukin17,IL17)、转化生长因子β_1(Transforming growth factor-β_1,TGF-β_1)的表达。结果对照组及八正散高剂量组大鼠膀胱黏膜固有层炎细胞浸润、间质水肿、局灶性坏死和黏膜固有层内Brunn巢及腺上皮化生等较模型组明显减少。与空白组比较,模型组的IL-6、IL-17和TGF-β_1的含量显著升高,差异有统计学意义(P<0.01);与模型组比较,对照组及高剂量八正散组IL-6、IL-17和TGF-β_1的含量显著降低,差异有统计学意义(P<0.05)。中剂量组1L-6、TGF-β_1的含量显著降低,差异有统计学意义(P<0.05)。结论八正散可以有效改善腺性膀胱炎模型大鼠膀胱病变程度,推测与下调IL-6、IL-17和TGF-β_1含量有关。
Objective To investigate the mechanism of Bazheng San on cytokines in rats with pathological glandular cystitis. Methods The model rats with glandular cystitis were randomly divided into blank group, model group, control group and different doses of Bazheng San (high, medium and low) groups. The control group was given hydroxycamptothecin 3mg / kg intravesical instillation, eight positive and negative high, medium and low dose groups were given the same volume of eight are bulk liquid (crude drug concentration of 0.84g / mL, 0.42g / mL, 0.2lg / mL) twice daily, perfusion / gavage for 2 consecutive weeks. The pathological changes of bladder were observed by HE staining. The expression of interleukin 6 (IL-6), interleukin 17 (IL17) and transforming growth factor-β 1 (TGF-β 1) in bladder tissue were detected by immunohistochemistry -β_1). Results Compared with the model group, the inflammatory cell infiltration, interstitial edema, focal necrosis and Brunn nest and glandular metaplasia in the lamina propria mucosa of control group and BZG high dose group were significantly decreased. Compared with the blank group, the contents of IL-6, IL-17 and TGF-β 1 in the model group were significantly increased (P <0.01). Compared with the model group, The levels of IL-6, IL-17 and TGF-β 1 were significantly decreased (P <0.05). The content of 1L-6 and TGF-β 1 in the middle-dose group was significantly lower than that in the middle-dose group (P <0.05). Conclusions Bazheng San can effectively improve the degree of bladder lesions in rats with glandular cystitis, which is presumed to be related to the decrease of IL-6, IL-17 and TGF-β 1 levels.
