应用MTT法测定SDH活性及肿瘤干细胞集落形成法对HOS-9603细胞系进行体外热灭活实验研究.结果表明:(1)45℃60min或50℃10min以上,HOS-8603基本失去细胞集落形成能力;(2)SDN的活性随温度升高和时间延长而下降;与细胞集落形成能力有较好的相关性,(3)提示了临床无需采用煮沸灭活即可将肿瘤细胞杀灭.
MTT assay was used to determine the activity of SDH and tumor stem cell colony formation assay. The heat inactivation of HOS-9603 cell line was studied in vitro. The results showed that: (1) At 45°C for 60 minutes or 50°C for more than 10 minutes, HOS-8603 basically lost the ability to form colonies; (2) SDN activity decreased with increasing temperature and time; it had better ability to form colonies. Relevance, (3) suggests that clinically it is not necessary to use boiled inactivation to kill tumor cells.
