目的:体外无血清条件下诱导人脐带间充质干细胞向肝样细胞分化。方法:在无血清条件培养体系下,利用肝细胞生长因子和致瘤素联合诱导人脐带间充质干细胞向肝样细胞分化。观察细胞形态学变化,检测诱导细胞甲胎蛋白、细胞角蛋白-18、白蛋白mRNA及相应蛋白的表达及糖原储存情况。结果:经肝细胞生长因子、致瘤素联合诱导后,细胞形态由长梭形变为类圆形,表达甲胎蛋白、细胞角蛋白-18、白蛋白mRNA及相应蛋白,具有合成和储存糖元的功能。结论:体外无血清培养扩增人脐带间充质干细胞,经肝细胞生长因子(20ng/ml)与致瘤素(20ng/ml)联合诱导后,初步具有向肝样细胞分化的能力。
OBJECTIVE: To induce human umbilical cord mesenchymal stem cells to differentiate into hepatocyte-like cells under serum-free conditions. Methods: Human umbilical cord-derived mesenchymal stem cells were induced to differentiate into hepatocyte-like cells under the condition of serum-free culture system by using combination of hepatocyte growth factor and oncostatin. The morphological changes of cells were observed, and the expression of alpha-fetoprotein, cytokeratin-18, albumin mRNA and corresponding proteins and the storage of glycogen were detected. Results: After induction by hepatocyte growth factor and tumorigenin, the cell morphology changed from long spindle to round, expressing alpha-fetoprotein, cytokeratin-18, albumin mRNA and corresponding proteins with the ability to synthesize and store glycogen Function CONCLUSION: Human umbilical cord mesenchymal stem cells (MSCs) were expanded in serum-free medium in vitro and initially showed the ability to differentiate into hepatocyte-like cells when induced by 20ng / ml hepatocyte growth factor (20ng / ml).
