目的观察异丙酚对兔脑蓝斑去甲肾上腺素(NE)代谢的影响,探讨异丙酚引起循环抑制的中枢机制。方法选择健康雄性新西兰兔9只,静脉输注异丙酚前后,使用推挽灌流法收集蓝斑区灌流样本,高效液相色谱法测定NE浓度。结果静脉注射异丙酚负荷量2mg/kg及维持量150μg·kg-1·min-1后,脑灌流液中NE浓度均有下降(P<0.01),在使用负荷量后10min内下降幅度最大,降幅达75.5%[从(15.9±3.2)pg/μl下降到(3.9±0.5)pg/μl]。结论异丙酚可降低蓝斑区NE的浓度,可能是其引起循环抑制的中枢机制之一。
Objective To investigate the effect of propofol on the metabolism of noradrenaline (NE) in the brain and to explore the central mechanism of propofol-induced circulatory depression. Methods Nine male New Zealand white rabbits were selected. Before and after intravenous infusion of propofol, the samples of locus coeruleus were collected by push-pull perfusion method and NE concentration was determined by high performance liquid chromatography. Results After intravenous injection of propofol at a dose of 2 mg / kg and a maintenance dose of 150 μg · kg-1 · min-1, the concentration of NE in the brain perfusion fluid decreased (P <0.01), and decreased most significantly within 10 min , A decrease of 75.5% [from (15.9 ± 3.2) pg / μl to (3.9 ± 0.5) pg / μl]. Conclusion Propofol can reduce the concentration of NE in locus coeruleus and may be one of the central mechanisms of its inhibition.
