目的:探讨厚朴酚(mangolol,Mag)对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱导PC12细胞凋亡的抑制作用及可能作用机制。方法:将厚朴酚(终浓度0.1,10μmol.L-1)或/和MPTP(终浓度50,100,150,200μmol.L-1)加入到培养的PC12细胞中。四甲基偶氮唑盐法(MTT法)检测细胞增殖活性,用流式细胞仪检测细胞凋亡率,以及Western-blot法检测Bax和Bcl-2的蛋白表达变化。结果:在加入不同浓度的MPTP处理细胞24 h,细胞增殖活性渐次降低,而厚朴酚0.1~10μmol.L-1预处理1 h可明显减轻MPTP导致的PC12细胞的损伤,抑制细胞凋亡,以及Bcl-2/Bax比值的改变。结论:厚朴酚对MPTP诱导的PC12细胞凋亡损伤有一定的抑制作用,其作用机制涉及到影响细胞内Bcl-2/Bax蛋白的表达。
AIM: To investigate the inhibitory effect of mangolol (Mag) on the apoptosis of PC12 cells induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and its possible mechanism. Methods: Magnolol (final concentration 0.1, 10 μmol·L-1) or / and MPTP (final concentration 50, 100, 150, 200 μmol·L-1) were added to cultured PC12 cells. MTT assay was used to detect cell proliferation. Flow cytometry was used to detect the apoptosis rate. The protein expression of Bax and Bcl-2 was detected by Western-blot. RESULTS: After adding MPTP for 24 h, the proliferative activity of MPTP decreased gradually. When pretreatment with magnolol 0.1 ~ 10 μmol·L-1 for 1 h, MPTP could significantly reduce the damage of PC12 cells and inhibit the apoptosis. And the change of Bcl-2 / Bax ratio. CONCLUSION: Magnolol can inhibit the apoptosis of PC12 cells induced by MPTP to a certain extent. Its mechanism of action involves in affecting the expression of Bcl-2 / Bax protein.
