目的:探讨粉防己碱与奈达铂联合应用对人肝癌细胞株7402的影响及其作用机制。方法:经不同浓度的粉防己碱和(或)奈达铂处理7402细胞后,采用MTT法测定细胞的增殖抑制效应;AO/EB荧光染色、流式细胞术观察细胞凋亡及细胞周期变化;免疫组织化学法观察凋亡相关基因表达的变化。结果:与粉防己碱或奈达铂单药作用相比,粉防己碱与奈达铂联合应用可显著增强人肝癌细胞增殖抑制作用和诱导肝癌细胞凋亡作用,联合用药后细胞周期分布发生改变,S期和G2/M期细胞比例下降,G0/G1期细胞比例上升,癌细胞Bc l-2基因表达明显下降,BAX表达显著增强。结论:粉防己碱与奈达铂联合应用具有显著协同抗肝癌作用,其机制可能与增强诱导肝癌细胞凋亡、细胞周期调控以及凋亡相关基因表达的调控有关。
Objective: To investigate the effect of tetrandrine combined with nedaplatin on human hepatoma cell line 7402 and its mechanism. Methods: The proliferation of 7402 cells treated with different concentrations of tetrandrine and / or nedaplatin were detected by MTT assay. The apoptosis and cell cycle were observed by AO / EB staining and flow cytometry. The changes of apoptosis related gene expression were observed by immunohistochemistry. Results: Compared with the tetrandrine or nedaplatin monotherapy, combination of tetrandrine and nedaplatin significantly enhanced the inhibition of human hepatocellular carcinoma cell proliferation and induced the apoptosis of hepatocellular carcinoma cells, and the cell cycle distribution changed after combined treatment , The proportion of cells in S phase and G2 / M phase decreased, the proportion of cells in G0 / G1 phase increased, the expression of Bcl-2 gene in cancer cells decreased significantly, and the expression of BAX increased significantly. Conclusion: Combination of tetrandrine and nedaplatin has a significant synergistic effect against HCC. The mechanism may be related to the enhancement of apoptosis induction, cell cycle regulation and the expression of apoptosis related genes in hepatocellular carcinoma cells.
