莪术挥发油(1∶10甲醇溶液)在硅胶G薄层上,用石油醚(30~60℃)-丙酮-乙酸乙酯(94∶5∶1)展开,碘蒸气显色两分钟,挥去碘后,将莪术醇斑点刮至具塞刻度离心管中,加入2%间苯三酚乙醇溶液1.6ml,放置并振摇约20分钟,继加浓盐酸至总体积为5.0m1,摇匀,离心数分钟后倾取上清液以试剂空白为对照于520nm测定。因间苯三酚与莪术醇显色反应的专属性较强,灵敏度亦较高,且莪术挥发油中各组分采用上述层离条件可有效地分离,故本法的测定结果比较稳定,操作亦甚简便。
Curcuma zedoaria volatile oil (1:10 methanol solution) was applied to a thin layer of silica gel G and developed with petroleum ether (30-60°C)-acetone-ethyl acetate (94:5:1). The iodine vapor was developed for two minutes and the iodine was evaporated. Afterwards, the curcumin alcohol spot was scraped into a stoppered centrifuge tube, 2% pyrogallol ethanol solution (1.6 ml) was added, placed and shaken for about 20 minutes, followed by adding concentrated hydrochloric acid to a total volume of 5.0 ml, shaken, and centrifuged. After a few minutes the supernatant was decanted and the reagent blank was determined as a control at 520 nm. Due to the specificity of the color reaction of phloroglucinol and curcumol, the sensitivity is also high, and the components of Curcuma zedoaria volatile oil can be effectively separated by the above-mentioned delamination conditions. Therefore, the assay result of this method is relatively stable, and the operation is also stable. Very simple.
