目的:优选黄独总皂苷的分离纯化工艺。方法:以黄独总皂苷含量为指标,采用紫外分光光度法测定其含量,动态吸附-解吸法筛选大孔树脂型号,并优选黄独总皂苷的分离纯化工艺。结果:D-101型大孔树脂富集纯化黄独总皂苷的最佳工艺为黄独提取液质量浓度0.2 g.mL-1,上样量6 BV,流速1.0 mL.min-1,加4 BV水洗除杂,50%乙醇4 BV解吸,收集洗脱液。此工艺条件下所得提取物黄独总皂苷质量分数61.85%。结论:该优选工艺简单可行,能满足大生产实际要求。
Objective: To optimize the isolation and purification of total saponins from Huangdu. Methods: The total saponin content of yellow sago as an indicator, the content was determined by UV spectrophotometry, dynamic adsorption - desorption method for screening macroporous resin model, and the optimization of the total saponin Purification Technology. Results: The optimum conditions for the enrichment and purification of total saponins from Pseudostellaria tabacum by D-101 macroporous resin were 0.2 g.mL-1, 6 BV, 1.0 mL.min-1 and 4 BV washing impurity, 50% ethanol 4 BV desorption, collecting eluent. Under the condition of the process, the total mass fraction of the total saponin of the yellow saponin is 61.85%. Conclusion: The optimal process is simple and feasible, to meet the actual production requirements.
