目的 从采自云南省中甸县的中华山荷叶 (DiphylleiasinensisLi)的根状茎中分离出可产生鬼臼毒素或其类似物的内生真菌。方法 按常规无菌方法分离内生真菌 ,纯化后接种于PDA斜面备用 ,液体培养基为改良的MM培养基。用TLC和HPLC方法对发酵产物进行检测。结果 经TLC检测 ,发现 99SJ2 1的Rf值与标样的Rf值相同。经HPLC分析 ,发现 99SJ2 1的发酵产物在相同的设定条件下与标样在tR4 34min的波峰一致。经鉴定 ,99SJ2 1为纠缠青霉 (PenicilliumimplicatumBiourage)。 结论 99SJ2 1的确产鬼臼毒素类化合物
Objective To isolate endophytic fungi that produce podophyllotoxin or its analogues from the rhizome of Diphylleiasinensis Li collected from Zhongdian County, Yunnan Province. Methods The endophytic fungi were isolated by routine aseptic method, purified and inoculated on PDA slant for use. The liquid culture medium was modified MM medium. The fermentation product was detected by TLC and HPLC methods. Results After TLC detection, the Rf value of 99SJ2 1 was found to be the same as the standard Rf value. After HPLC analysis, it was found that the 99SJ21 fermented product was consistent with the standard at tR4 34min peak under the same set conditions. After identification, 99SJ2 1 is Penicillium implicatum Biourage. Conclusion 99SJ21 does produce podophyllotoxins
