目的探讨环腺苷酸(cAMP)类似物8-溴-环腺苷酸(8-Br-cAMP)对人原代培养的膀胱癌细胞中c-erbB-2、c-myc蛋白及mRNA表达的影响。方法原代培养膀胱癌细胞,采用完整细胞免疫组化、原位杂交方法检测膀胱癌细胞中c-erbB-2、c-myc蛋白及mRNA的表达。结果8-Br-cAMP处理后的膀胱癌细胞中c-erbB-2、c-myc蛋白及mRNA表达水平明显低于未处理组,两组相比差异均有统计学意义(P<0.05)。结论8-Br-cAMP可抑制膀胱癌细胞中c-erbB-2、c-myc蛋白及mRNA表达,进而抑制肿瘤细胞的分裂和增殖。
Objective To investigate the expression of c-erbB-2, c-myc protein and mRNA in human primary cultured bladder cancer cells treated with 8-Br-cAMP, an analogue of cyclic adenosine monophosphate (cAMP) influences. Methods Primary bladder cancer cells were cultured. The expression of c-erbB-2, c-myc protein and mRNA in bladder cancer cells were detected by whole cell immunohistochemistry and in situ hybridization. Results The expression of c-erbB-2, c-myc protein and mRNA in 8-Br-cAMP-treated bladder cancer cells was significantly lower than that in the untreated group. There was significant difference between the two groups (P <0.05). Conclusion 8-Br-cAMP can inhibit the expression of c-erbB-2, c-myc protein and mRNA in bladder cancer cells, and then inhibit tumor cell division and proliferation.
