目的:测定宁夏沙棘果实中异鼠李素的含量。方法:采用HPLC法,色谱柱为C18-柱,流动相为甲醇-醋酸缓冲液(pH=4.86)(52∶48),流速为1.0mL.min-1;检测波长370nm,柱温30℃。结果:异鼠李素检测浓度在0.03~0.4μgmL-1范围内,具有良好的线性关系(R=0.9994)。平均加样回收率为99.46%(RSD为0.53%,n=5)。结论:沙棘浸膏中含异鼠李素26.2mg.g-1,生药中的异鼠李素含量为11.8mg.g-1。
Objective: To determine the content of isorhamnetin in sea buckthorn in Ningxia. Methods: The HPLC method was adopted. The column was C18-column. The mobile phase consisted of methanol-acetic acid buffer (pH = 4.86) (52:48) and the flow rate was 1.0 mL.min-1. The detection wavelength was 370 nm and the column temperature was 30 ℃. Results: Isorhamnetin has a good linearity (R = 0.9994) in the range of 0.03-0.4 μg mL-1. The average recovery was 99.46% (RSD 0.53%, n = 5). CONCLUSION: Isofulucin 26.2 mg.g-1 is contained in seabuckthorn extract and isorhamnetin is 11.8 mg.g-1 in crude drug.
