目的:探讨舒心祛风汤对大鼠动脉粥样硬化形成的影响。方法:60只SD大鼠随机分成空白组、模型组、舒心祛风汤小剂量组、舒心祛风汤大剂量组、舒降之组,每组12只。建立动脉粥样硬化大鼠模型。8周后检测血脂、血管紧张素Ⅰ、血管紧张素Ⅱ的含量;观察胸主动脉内膜的形态学,采用免疫组织化学法检测单核细胞趋化蛋白-1(MCP-1)、P-选择素(Ps)的表达,RT-PCR法检测MCP-1、Ps的mRNA表达。结果:舒心祛风汤能降低血低密度脂蛋白,升高血高密度脂蛋白;降低血管紧张素Ⅱ水平;调节血小板活化和趋化因子MCP-1的蛋白表达;减少MCP-1和Ps mRNA在转录水平上的过度表达。结论:舒心祛风汤对动脉粥样硬化有一定的预防作用,其机制可能与减少血小板活化和抑制炎症因子的表达有关。
Objective: To investigate the effect of Shuxin Qufeng Decoction on the formation of atherosclerosis in rats. Methods: Sixty SD rats were randomly divided into blank group, model group, Shuxin Qufeng Decoction small dose group, Shuxin Qufeng Decoction large dose group, and Shujiangzhi group, 12 in each group. Establish a rat model of atherosclerosis. Serum lipids, angiotensin I, and angiotensin II levels were measured after 8 weeks. Morphology of thoracic aorta intima was observed. Monocyte chemoattractant protein-1 (MCP-1) and P-protein were detected by immunohistochemistry. The expression of Ps was detected by RT-PCR and mRNA expression of MCP-1 and Ps was detected. Results: Shuxin Qufeng Decoction can lower blood low-density lipoprotein, increase blood high-density lipoprotein; reduce the level of angiotensin II; regulate platelet activation and protein expression of chemokine MCP-1; reduce MCP-1 and Ps mRNA Overexpression at the transcriptional level. Conclusion: Shuxin Qufeng Decoction has a certain preventive effect on atherosclerosis. The mechanism may be related to reducing platelet activation and inhibiting the expression of inflammatory factors.
