目的:建立桑叶中芦丁含量的测定方法。方法:高效液相色谱法,以VARIAN ODS(250 mm×4.6 mm,5μm)为色谱柱,乙腈(A)-0.5%磷酸(B)做流动相,经梯度洗脱[0.01 min,A-B(18∶82);4 min,A-B(21∶79);14 min,A-B(8∶92);17 min,A-B(18∶82);18 min,A-B(18∶82)],流速1.0 ml/min,检测波长为358 nm,柱温为40℃,紫外检测器检测。结果:芦丁浓度5μg/ml~100μg/ml范围内呈良好的线性关系,(r=0.99996,n=5),精确度(RSD=1.9%~2.1%),平均回收率试验为102%。最低检出限为0.1μg/ml(以进量浓度计)。结论:实验结果表明该方法灵敏度高,精密度和准确度好,是一种简便、快速分析的方法。
Objective: To establish a method for the determination of rutin in mulberry leaves.
