目的观察清热除痹方对佐剂性关节炎(adjuvant arthritis,AA)大鼠的抗炎消肿作用及对白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的影响。方法应用弗氏完全佐剂诱导大鼠AA模型。实验分清热除痹方高、中、低剂量组(22.4,11.2,5.6 g·kg-1),美洛昔康组(0.746 g·kg-1)、模型组及空白对照组。实验第11天开始给药,连续10 d。分别于实验第10天、第14天、第18天、第20天,测量关节肿胀程度及关节炎指数。末次给药24 h后,采用酶联免疫试验(ELISA)检测大鼠关节滑膜匀浆中IL-1β、TNF-α的含量。结果清热除痹方各组在实验第18天开始大鼠足肿胀度明显低于模型组(P<0.05);第14天开始关节炎指数明显低于模型组(P<0.05);与模型组相比,清热除痹方各组IL-1β、TNF-α含量明显下降(P<0.05),且各实验结果均有一定的剂量依赖性。结论清热除痹方可明显抑制模型大鼠足肿胀度及关节炎指数,具有一定抗炎消肿作用。降低关节滑膜匀浆中IL-1β、TNF-α的含量可能是清热除痹方抗炎消肿作用的机理之一。
Objective To observe the anti-inflammatory and detumescence effects of Qingre Qiebi Recipe on adjuvant arthritis (AA) rats and the effect on the expression of interleukin-1β (IL-1β), tumor necrosis factor-α influences. Methods The AA model was induced by Freund’s complete adjuvant. The experimental group was divided into two groups: high, medium and low dose of Pycnogenol group (22.4,11.2,5.6 g · kg -1), meloxicam group (0.746 g · kg -1), model group and blank control group. The experiment began on the 11th day for 10 days. The degree of joint swelling and arthritis index were measured on the 10th day, the 14th day, the 18th day and the 20th day respectively. After the last administration for 24 h, the contents of IL-1β and TNF-α in synovial homogenate of rats were detected by enzyme-linked immunosorbent assay (ELISA). Results The degree of paw edema in the Qingre Chiebi group was significantly lower than that in the untreated group on the 18th day (P <0.05), and the arthritis index was lower on the 14th day (P <0.05). Compared with the model group Compared with other groups, the content of IL-1β and TNF-α of Qingre Qixifang group were significantly decreased (P <0.05), and the experimental results were dose-dependent. Conclusion Qingre Qubi Fang can significantly inhibit the rat model of foot swelling and arthritis index, has a certain degree of anti-inflammatory swelling. Reducing the content of IL-1β and TNF-α in synovial homogenate may be one of the mechanisms of anti-inflammatory and detumescence effects of Qingre Zhibi.
