目的观察醒脑静微乳经鼻给药后对大鼠行为及鼻黏膜形态学的影响,探索开发新型复方制剂。方法健康SD大鼠125只,随机分为空白微乳组、空白溶剂组、醒脑静微乳组(载药量为84.4g/L)、醒脑静滴鼻液组(载药量为84.4 g/L)以及生理盐水组,每组25只,各组每日给药3次,每次每鼻孔定量注入50μL相应液体,连续给药7 d,于给药1、4、7 d和停药1、2周后,取鼻中隔黏膜,制备石蜡切片,HE染色,光镜下观察,并对大鼠进行行为学观察。结果各组鼻腔给药后,大鼠有不同程度的行为反应。切片观察可知空白微乳组、空白溶剂组与醒脑静微乳组给药7 d后对大鼠鼻黏膜有一定的刺激性,且刺激性相当(P>0.05),但损伤于停药1周后即恢复正常;醒脑静滴鼻液组给药7 d后对大鼠鼻黏膜刺激性显著高于微乳组(P<0.05)。结论醒脑静中成分对大鼠鼻黏膜有较大的刺激性,但将其制成微乳后可减少药物对鼻黏膜的刺激性且损伤于1周后恢复正常。
Objective To observe the effects of xingnaojing microemulsion on nasal mucosa morphology and nasal mucosa nasal administration, and explore the development of new compound preparations. Methods 125 healthy SD rats were randomly divided into two groups: blank microemulsion group, blank solvent group, Xingnaojing microemulsion group (drug loading 84.4g / L), Xingnaojing nasal drops group (drug loading 84.4 g / L) and saline group, 25 rats in each group. Slice observation showed that the blank microemulsion group, blank solvent group and Xingjingjing microemulsion group had certain irritation and irritation to the nasal mucosa for 7 d after administration (P> 0.05), but the injury was stopped at 1 The rats in Xingnongjing nasal drops group were significantly irritated to the nasal mucosa 7 days after administration (P <0.05). Conclusion Xingnao Jingzhong ingredient has a greater irritating effect on the nasal mucosa of rats. However, when it is made into microemulsion, it can reduce the irritation of the nasal mucosa and the damage resumes after 1 week.
