目的 研究BcL 2、BAX基因与急性白血病的关系及其临床意义。方法 采用逆转录多聚酶链式反应技术 ,对 36例急性白血病患儿及 10例血小板减少性紫癜患儿骨髓单个核细胞进行BcL 2、BAX基因表达的检测。结果 ①BcL 2基因的表达在初发组、复发组高于对照组 ,复发组高于初发组和完全缓解组 ,差异有显著性 ;完全缓解组和对照组无显著性差异。BAX基因的表达在初发组、复发组、完全缓解组和对照组相比差异均无显著性。②BcL 2、BAX基因的表达水平与性别、年龄、白细胞数、免疫表型、骨髓中幼稚细胞百分数和肝脾淋巴结肿大均无相关性。结论 白血病的发生可能与BcL 2基因的高表达有关 ;BcL 2高表达者易复发 ,BcL 2可作为估计预后的指标之一
Objective To study the relationship between Bcl-2, BAX gene and acute leukemia and its clinical significance. Methods BcL 2 and BAX gene expression in bone marrow mononuclear cells of 36 children with acute leukemia and 10 children with thrombocytopenic purpura were detected by reverse transcription polymerase chain reaction. Results ① The expression of Bcl-2 gene was higher in the primary group and the recurrent group than in the control group. The recurrence group was significantly higher than the initial group and the complete remission group. There was no significant difference between the complete remission group and the control group. BAX gene expression in the initial group, recurrence group, complete remission group compared with the control group, no significant difference. ② There was no correlation between the expression of Bcl-2 and BAX genes in gender, age, leukocyte count, immunophenotype, percentage of immature cells in bone marrow and hepatosplenomegaly. Conclusions The occurrence of leukemia may be related to the high expression of BcL 2 gene. The high expression of BcL 2 is easy to relapse and BcL 2 may be used as one of the prognostic indicators
