[目的]探讨鹤草酚对人肺腺癌A549细胞株的放射增敏作用。[方法]MTT法测定0、0.5、1.0、2.0、4.0、8.0、16.0和32.0μmol/L浓度鹤草酚对人肺腺癌A549细胞株的生长抑制率。实验分4组:常氧照射组、乏氧照射组、0.887μmol/L鹤草酚+乏氧照射组和1.774μmol/L鹤草酚+乏氧照射组,测定各组存活分数(SF),计算细胞生存率,拟合细胞生存曲线并计算放射增敏比。[结果]MTT结果表明,鹤草酚对人肺腺癌A549细胞有生长抑制作用,不同浓度鹤草酚作用组间细胞抑制率有显著性差异(F=26.818,P=0.002)。鹤草酚作用A549细胞的IC50为8.87μmol/L。0.887、1.774μmol/L鹤草酚联合乏氧照射组的D0、Dq和2Gy时的细胞存活分数(SF2)均小于单纯乏氧照射组。0.887μmol/L和1.774μmol/L鹤草酚联合乏氧照射组的放射增敏比(SER)分别为1.484和1.525。[结论]鹤草酚对人肺腺癌A549细胞株有抑制增殖作用,其常氧培养IC50值为8.87μmol/L。鹤草酚对乏氧培养的人肺腺癌A549细胞株有放射增敏作用,随着鹤草酚浓度的增加其放射增敏作用增强。
[Objective] To investigate the radiosensitization effect of oxybenzene on human lung adenocarcinoma A549 cell line. [Methods] The growth inhibition rate of human lung adenocarcinoma A549 cell line with 0, 0.5, 1.0, 2.0, 4.0, 8.0, 16.0 and 32.0 μmol / L concentration of oxybenzene was determined by MTT assay. The experiment was divided into four groups: normoxia group, hypoxia group, 0.887μmol / L cilly-phenol-hypoxic group and 1.774μmol / Calculate the cell survival rate, fit the cell survival curve and calculate the radiosensitization ratio. [Result] The results of MTT showed that the growth inhibition of human lung adenocarcinoma A549 cells was induced by oxybenzone, and the cell inhibition rate was significantly different between groups (F = 26.818, P = 0.002). The IC50 of A549 cells with oxycodone was 8.87μmol / L. The cell viability fraction (SF2) at D0, Dq and 2Gy of 0.887 and 1.774μmol / L was significantly lower than that of hypoxia alone group. The radiosensitization ratio (SER) of 0.887μmol / L and 1.774μmol / L of the combination of oxybenzone and hypoxia group were 1.484 and 1.525, respectively. [Conclusion] Ecgonimal could inhibit the proliferation of human lung adenocarcinoma A549 cell line with IC50 value of 8.87μmol / L. Ciclopirox has radiosensitization effect on hypoxia-cultured human lung adenocarcinoma A549 cell line, and its radiosensitization effect is enhanced with the increase of its concentration.
