目的研究促红细胞生成素(EPO)对高糖培养小鼠肾脏足细胞肾素受体(RR)表达的影响。方法体外培养和分化小鼠肾小球足细胞,按培养条件不同分为正常对照组(Con,含5.6mmol/L葡萄糖),高渗组(含5.6mmol/L葡萄糖+24.4mmol/L甘露醇),高糖1组(含15mmol/L葡萄糖),高糖2组(含30mmol/L葡萄糖)。每个培养条件分别以0、10、20、50U/ml的EPO进行干预,干预24h后收集足细胞。RT-PCR、Western blot检测RR表达情况。结果不同葡萄糖浓度处理后,高糖1组、高糖2组足细胞RR mRNA和蛋白表达相对量与Con组比较,差异有统计学意义(P<0.05)。不同EPO浓度干预各组足细胞24h后,各组均在10U/ml EPO浓度时,足细胞RR mRNA和蛋白表达最低,与各组不加EPO干预比较,差异有统计学意义(P<0.05)。结论 EPO对高糖诱导的足细胞RR mRNA及蛋白过表达有调节作用。
Objective To investigate the effect of erythropoietin (EPO) on renin receptor (RR) expression in renal podocytes of high glucose-cultured mice. Methods Glomerular podocytes were cultured and differentiated in vitro and divided into normal control group (Con containing 5.6mmol / L glucose) and hypertonic group (containing 5.6mmol / L glucose + 24.4mmol / L mannitol ), High glucose group 1 (containing 15mmol / L glucose), high glucose group 2 (containing 30mmol / L glucose). Each culture conditions were 0,10,20,50U / ml of EPO intervention, 24 hours after the intervention of podocyte collection. RT-PCR, Western blot detection of RR expression. Results After treatment with different glucose concentrations, the relative amounts of RR mRNA and protein expression in podocyte of high glucose group 1 and high glucose group 2 were significantly different from that of Con group (P <0.05). After intervention with EPO at different concentrations for 24 hours, the expressions of RR mRNA and protein in podocytes were the lowest at 10 U / ml EPO, and there was significant difference between them (P <0.05) . Conclusion EPO can regulate RR mRNA and protein overexpression in podocytes induced by high glucose.
