目的探讨Skp2和p27在乳腺癌分子亚型中的表达及与基底样型乳腺癌的相关性。方法 1 452例乳腺癌标本制作组织芯片,免疫组化MaxVision快捷法检测ER、PR、HER2、CK5/6和EGFR,按照Nielsen标准进行分子分型。同时检测Skp2和p27。结果在乳腺癌基底样型、HER2过表达、裸表达型、腺腔A型、腺腔B型中,Skp2表达率为63.3%、51.9%、53.1%、21.3%和13.7%;p27表达率为35.2%、46.3%、41.4%、55.9%和63.0%。Skp2在基底样型、HER2过表达型和裸表达型的表达中显著高于腺腔A和B型(P<0.01);而p27在基底样型、HER2过表达型和裸表达型中的表达率显著低于腺腔A和B型(P<0.05)。Skp2和p27的表达水平与ER表达相关(P<0.01)。Skp2和p27的表达在基底样型乳腺癌中呈负相关(P<0.01)。结论 Skp2的高表达及p27的失表达可能与ER阴性乳腺癌的发生和发展有关,并且Skp2在基底样型乳腺癌的发生、发展中起重要作用。在基底样型乳腺癌中二者基因相关性的研究为其发病机制的阐明提供帮助。
Objective To investigate the expression of Skp2 and p27 in breast cancer molecular subtypes and their relationship with basal-like breast cancer. Methods A total of 452 breast cancer specimens were made into tissue microarrays. The MaxVision rapid immunohistochemical method was used to detect ER, PR, HER2, CK5 / 6 and EGFR. The molecular typing was performed according to the Nielsen standard. Skp2 and p27 were also tested. Results Skp2 expression rates were 63.3%, 51.9%, 53.1%, 21.3% and 13.7% in breast cancer basal, HER2 overexpression, naked expression, glandular cavity type A and glandular cavity type B, respectively. 35.2%, 46.3%, 41.4%, 55.9% and 63.0%. The expression of Skp2 in basal-type, HER2 overexpression and overexpression was significantly higher than that in adenomas A and B (P <0.01), while the expression of p27 in basal-type, HER2 overexpression and overexpression Rate was significantly lower than the gland cavity A and B type (P <0.05). Conclusion The high expression of Skp2 and the loss of p27 expression may be related to the occurrence and development of ER-negative breast cancer, and Skp2 plays an important role in the occurrence and development of basal-like breast cancer. In the basal-like breast cancer gene-related research for the clarification of its pathogenesis to help.
