采用超声技术提取中药刺五加中的乌索酸和齐墩果酸,并建立HPLC-PAD法测定了乌索酸和齐墩果酸的含量。色谱柱为Nova-Pak C18(3.9 mm×300 mm,4μm),流动相为甲醇∶水(体积比,88∶12),流速为0.8 ml/min,二极管阵列检测器检测,检测波长210 nm,柱温25℃。乌索酸线性范围为0.136~1.224μg,线性回归系数为0.999 5;齐墩果酸线性范围为0.052~0.468μg,线性回归系数为0.999 4。样品中乌索酸的平均回收率为98.1%,RSD为1.1%(n=5);齐墩果酸的平均回收率为97.5%,RSD为1.0%(n=5)。该方法操作简便,结果准确、可靠,可用于刺五加中乌索酸和齐墩果酸含量的测定分析。
Ultrasonic technique was used to extract ursolic acid and oleanolic acid from Acanthopanax senticosus, and the HPLC-PAD method was used to determine the content of ursolic acid and oleanolic acid. The column was Nova-Pak C18 (3.9 mm×300 mm, 4 μm), the mobile phase was methanol:water (volume ratio, 88:12), the flow rate was 0.8 ml/min, diode array detector was used, detection wavelength was 210 nm, Column temperature 25°C. The linear range of ursolic acid is 0.136~1.224μg, the linear regression coefficient is 0.999 5, the linear range of oleanolic acid is 0.052~0.468μg, and the linear regression coefficient is 0.999 4. The average recovery of ursolic acid in the sample was 98.1%, RSD was 1.1% (n=5), the average recovery of oleanolic acid was 97.5%, and the RSD was 1.0% (n=5). The method is simple, accurate and reliable. It can be used for the determination of ursolic acid and oleanolic acid in Acanthopanax senticosus.
