观察复方 86 1含药血清对HSC T6细胞Ⅰ型胶原 (CoL Ⅰ )、Ⅲ型胶原 (CoL Ⅲ )mRNA表达影响。以不同剂量复方 86 1灌胃大鼠制备的含药血清 ,作用HSC T6细胞 4 8h ,应用逆转录定量PCR方法测定其对HSC T6细胞CoL Ⅰ、CoL ⅢmRNA表达的影响。结果表明 :复方 86 10 5、1.0、2 .0、6 .0g kg等不同剂量含药血清HSC T6细胞CoL Ⅰ、CoL ⅢmRNA表达。与空白对照组比较 ,具有显著性 (P <0 .0 5或P ,0 .0 1)。因此 ,复方 86 1可降低HSC T6细胞CoL Ⅰ、CoL ⅢmRNA表达水平 ,具有抗肝纤维化作用。
Observed the effects of compound 86 1 drug-containing serum on the expression of type I collagen (CoL I) and type III collagen (CoL III) mRNA in HSC T6 cells. The drug-containing serum prepared by intragastric administration of different doses of compound 86 1 was used to treat HSC T6 cells for 48 hours. The effect of HSC T6 cells on expression of CoL I and CoH III mRNA was determined by reverse transcription quantitative PCR. The results showed that the mRNA expression of CoL I and CoL III in HSC T6 cells with different doses of 8610 5, 1.0, 2.0, and 6.0 g kg. Compared with the blank control group, it was significant (P <0.05 or P0, 0.01). Therefore, compound 86 1 can reduce the expression of CoL I and CoL III mRNA in HSC T6 cells, and has anti-fibrotic effect.
