目的:研究调节性T细胞(Treg/Th17)细胞失衡在巨细胞病毒感染(CMV)免疫调节机制中的作用。方法:将CMV感染患儿按诊断标准分为激活感染组与潜伏感染组,同时设立正常对照组,采用流式细胞术(FCM)分析各组外周血Treg、Th17的百分比,并计算Treg/Th17比值;同时采用ELISA和RT-PCR法检测Treg主要相关因子(IL-10、Foxp3)和Th17主要相关因子(TGF-β、IL-17、IL-6、IL-23及ROR-γt)表达水平。结果:与对照组比较,CMV感染后Treg细胞百分率降低,Th17细胞升高,致Treg/Th17比值下降(P<0.05);CMV感染后两组间比较,激活感染组Treg/Th17比值和Treg主要相关因子表达水平下降更明显,而Th17主要相关因子表达水平显著上调,差异均有统计学意义(P<0.05)。结论:Treg/Th17平衡参与了CMV感染发病免疫机制,并可能与病毒潜伏-激活状态相关。
AIM: To investigate the role of imbalance of regulatory T cell (Treg / Th17) cells in the immunomodulatory mechanism of cytomegalovirus infection (CMV). Methods: Children with CMV infection were divided into activated infection group and latent infection group according to the diagnostic criteria. At the same time, a normal control group was set up. The percentage of Treg and Th17 in each group were analyzed by flow cytometry (FCM) The expression of major related factors of Treg (IL-10, Foxp3) and Th17 (TGF-β, IL-17, IL-6, IL-23 and ROR-γt) were detected by ELISA and RT- . Results: Compared with the control group, the percentage of Treg cells decreased and Th17 cells increased, the ratio of Treg / Th17 decreased (P <0.05). After CMV infection, the ratio of Treg / Th17 in active infection group and Treg The expression of related factors decreased more significantly, while the expression levels of Th17 major related factors were significantly increased, the differences were statistically significant (P <0.05). CONCLUSION: The balance of Treg / Th17 is involved in the pathogenesis and pathogenesis of CMV infection and may be related to the latent-activation state of the virus.
