目的优化莲房原花青素提取纯化工艺并评价抗氧化活性。方法以正交设计L9(34)优化莲房原花青素提取工艺参数,用大孔吸附树脂进行纯化,而后通过1,1-二苯基-2-苦肼基(DPPH)和羟基自由基(·OH)清除率评价其体外抗氧化活性。结果在最佳提取工艺条件下(8倍量50%乙醇回流提取2次,每次3 h),提取率达91.3%,进一步通过大孔树脂AB-8纯化,最终提取物中原花青素纯度为80.7%。莲房原花青素抗氧化活性与葡萄籽原花青素无显著性差异,均明显优于维生素C。结论该提取纯化方法简单、高效,可用于莲房原花青素提取物的制备。莲房原花青素具有较强的抗氧化活性,值得进一步开发利用。
Objective To optimize the extraction and purification of proanthocyanidins in lotus seed and evaluate the antioxidant activity. Methods The orthogonal design L9 (34) was used to optimize the extraction parameters of proanthocyanidins in lotus seedlings and purified by macroporous adsorption resin. Then, DPPH and hydroxyl radical (· OH ) Clearance rate to evaluate its in vitro antioxidant activity. Results The extraction rate was 91.3% under the optimum extraction conditions (8 times the amount of 50% ethanol reflux extraction 3 times each time), and further purified by macroporous resin AB-8, the purity of procyanidin in the final extract was 80.7 %. Lotus proanthocyanidins antioxidant activity and grape seed proanthocyanidins no significant difference were significantly better than vitamin C. Conclusion The extraction and purification method is simple and efficient and can be used for the preparation of proanthocyanidins extract in lotus. Lotus proanthocyanidins with strong antioxidant activity, it is worth further development and utilization.
