目的:观察黑蒜提取液对大鼠心肌缺血再灌注(MRI)的干预作用,并探讨其作用的可能机制。方法:将20只Wistar大鼠随机分为假手术组、MRI组、黑蒜低、高剂量治疗组,分别给予0.9%氯化钠溶液及黑蒜提取液处理,实验结束后处死大鼠并腹主动脉抽血,检测大鼠心肌人髓过氧化物酶(MPO)活性,血清脂质氧化丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力,流式细胞仪检测心肌细胞凋亡,免疫组织化学法检测心肌细胞B淋巴细胞瘤2基因Bcl-2和Bcl-2相关X蛋白Bax蛋白表达水平的影响。结果:黑蒜提取液能够降低大鼠心肌MPO、血清MDA活性,提高血清SOD活力,减少大鼠心肌细胞凋亡(P<0.05,P<0.01),升高Bax的表达,降低Bcl-2表达。结论:黑蒜提取液能够增加对大鼠MRI的保护作用,其作用机制可能与减轻中性粒细胞(PMN)浸润,抑制脂质过氧化,提高抗氧化能力,通过升高Bax、降低Bcl-2表达减少心肌细胞凋亡有关。
Objective: To observe the intervention of black garlic extract on myocardial ischemia reperfusion (MRI) in rats and to explore its possible mechanism. Methods: Twenty Wistar rats were randomly divided into sham-operation group, MRI group, low-dose and high-dose black garlic group, treated with 0.9% sodium chloride solution and black garlic extract respectively. After the experiment, the rats were sacrificed Aorta blood was collected for determination of myocardial MPO activity, serum lipid peroxidation malondialdehyde (MDA) content, superoxide dismutase (SOD) activity, myocardial cell apoptosis by flow cytometry The expression of Bcl-2 and Bcl-2-related protein Bax in cardiomyocyte B lymphoblastoma 2 gene was detected by immunohistochemistry. Results: Black garlic extract could decrease the activity of MPO and serum MDA, increase the activity of serum SOD and decrease the cardiomyocyte apoptosis (P <0.05, P <0.01), increase the expression of Bax and decrease the expression of Bcl-2 . CONCLUSION: Black garlic extract can increase the protective effect of MRI on rats. The mechanism may be related to the reduction of PMN infiltration, the inhibition of lipid peroxidation and the enhancement of anti-oxidation ability. The increase of Bax and the decrease of Bcl- 2 expression in cardiomyocytes to reduce apoptosis.
