目的 探讨核心蛋白聚糖 (decorin)对正常肝细胞和肝星形细胞诱导凋亡的作用。方法 以正常人肝细胞株 (L 0 2细胞 )和大鼠肝星形细胞株 (HSC T6细胞 )为研究靶细胞 ,通过HE染色、流式细胞仪分析、TUNEL标记和电镜观察等方法证实decorin对肝细胞和肝星形细胞凋亡的诱导作用。结果 Decorin能够诱导两种靶细胞出现典型的凋亡细胞特征 ,细胞核浓缩、边聚 ,胞膜皱褶 ,凋亡小体形成。而且其诱导凋亡作用呈现剂量和时间效应。结论 Decorin能够诱导肝细胞和肝星形细胞凋亡 ,提示可以通过凋亡途径减少因损伤刺激而过度增生的细胞 ,以利于肝纤维化治疗
Objective To investigate the effect of decorin on apoptosis in normal hepatocytes and hepatic stellate cells. Methods Normal human hepatocyte cell line (L 0 2) and rat hepatic stellate cell line (HSC T6) were used as target cells. HE staining, flow cytometry analysis, TUNEL labeling and electron microscopy were used to confirm decorin Induction of apoptosis in hepatocytes and hepatic stellate cells. Results Decorin induced typical apoptotic cell characteristics in both target cells, including nucleus condensation, edge aggregation, cell membrane folds, and apoptotic body formation. And its induction of apoptosis showed dose and time effects. Conclusion Decorin can induce apoptosis in hepatocytes and hepatic stellate cells, suggesting that it can reduce the number of cells that are over-proliferated due to damage and stimulation in order to facilitate the treatment of liver fibrosis
