目的:观察甘草抗病毒有效部位(GD4)抑制呼吸道合胞病毒(RSV)作用。方法:采用煎煮法、二氯甲烷萃取法和柱层析法提取分离得到甘草抗病毒有效部位(GD4)。采用细胞病变抑制实验,观察GD4在Hela细胞中对RSV的抑制作用。结果:GD4半数中毒浓度(TC50)为0.23 mg/m l,抑制RSV的半数有效浓度(EC50)为28.73μg/m l,治疗指数(TI)为8.0;GD4对RSV的抑制作用存在量效关系;在感染后至少12 h以内给药,GD4能够有效地抑制RSV的复制。GD4不含有甘草酸。结论:甘草抗病毒有效部位(GD4)在Hela细胞中对呼吸道合胞病毒有明显的抑制作用。
OBJECTIVE: To observe the effect of licorice effective anti-viral site (GD4) on respiratory syncytial virus (RSV). Methods: The antiviral effective fraction (GD4) of Glycyrrhiza uralensis Fisch was extracted and extracted by boiling method, dichloromethane extraction method and column chromatography. The inhibition of RSV in Hela cells was observed using cytopathic inhibition assay. Results: The TC50 of GD4 was 0.23 mg/ml, the EC50 of RSV was 28.73 μg/ml, the therapeutic index (TI) was 8.0, and there was a dose-effect relationship between GD4 and RSV. Within at least 12 h after infection, GD4 can effectively inhibit the replication of RSV. GD4 does not contain glycyrrhizic acid. Conclusion: The effective anti-virus (GD4) site of Glycyrrhiza uralensis has obvious inhibitory effect on respiratory syncytial virus in Hela cells.
