目的动态观察大鼠实验性肝癌形成过程中抑凋谢基因bcl-2蛋白表达与γ-谷氨酰转肽酶(γ-GT)活性和谷胱甘肽硫转移酶(GST)活性改变的关系以及刺五加多糖的抑癌作用。方法用bcl-2单抗进行免疫组化SABC检测,用酶学动力学方法检测酶活性。结果饲喂致癌剂3-甲氨基偶氮苯(3′-MeDAB)的早期(15天),bcl-2蛋白表达已明显增高,实验结束时3′-MeDAB组阳性率为89.5%(17/19),刺五加组为64.7%(11/17)。bcl-2的过量表达与肝组织中γ-GT活性升高和GST活性降低相一致。结论bcl-2蛋白过度表达出现,对实验性肝癌的早期阶段,很可能在肿瘤的发生发展中起着促进作用。刺五加多糖对实验性肝癌的形成有一定抑制作用
Objective To observe the relationship between the expression of bcl-2 protein and the activity of γ-glutamyl transpeptidase (γ-GT) and the activity of glutathione-S-transferase (GST) in experimental rat liver cancer. Acanthopanax Spp. Polysaccharide inhibits cancer. Methods Immunohistochemical SABC assay was performed with bcl-2 monoclonal antibody and enzyme activity was measured by enzyme kinetics. Results In the early stage (15 days) of carcinogen 3-methylaminoazobenzene (3′-MeDAB), the expression of bcl-2 protein was significantly increased, and the positive rate in the 3′-MeDAB group was 89.5% at the end of the experiment ( 17/19), Acanthopanax group was 64.7% (11/17). Overexpression of bcl-2 is consistent with increased gamma-GT activity and decreased GST activity in liver tissue. Conclusion The overexpression of bcl-2 protein may play an important role in the development of tumors in the early stage of experimental liver cancer.
