本文旨在探讨从人尿中提取的生物制剂细胞分化剂(cell differential agent ⅡCDA-Ⅱ)诱导人肿瘤多药耐药细胞株KBV200细胞凋亡与逆转其多药耐药性的关系.主要采取的方法:应用四唑兰快速比色法(MTT法)检测细胞存活率,根据药物不同浓度及相关的细胞存活率,确定半数致死浓度,以了解CDA-Ⅱ逆转作用.应用形态学(瑞氏染色普通光镜观察及透射电镜观察),生化学(DNA末端转移标记法,TUNEL法)及流式细胞术分析细胞凋亡.结果发现CDA-Ⅱ毒性作用很低,浓度为2mg/ml时细胞存活率>90%,4mg/
The aim of this study was to investigate the relationship between apoptosis induced by cell differential agent II (CDA-II) extracted from human urine and the reversal of multidrug resistance in human tumor multidrug resistant cell line KBV200. Methods: Cell viability was measured by tetrazolium colorimetric assay (MTT method). The median lethal concentration was determined according to the concentration of the drug and the related cell survival rate to understand the reversal effect of CDA-II. Applied morphology (Rye’s staining Observation by ordinary light microscope and observation by transmission electron microscope), biochemistry (DNA terminal transfer labeling, TUNEL method) and flow cytometry analysis of cell apoptosis.
