目的确定犬细粒棘球绦虫感染环介导等温扩增技术(LAMP)最早检出日期。方法家犬8只,口服喂饲细粒棘球绦虫原头节,建立细粒棘球绦虫感染动物模型。感染后40d收集犬粪,用LAMP检测粪便中细粒棘球绦虫目的基因并与传统的PCR比较。结果 8只犬均获得细粒棘球绦虫感染,感染后第18dLAMP法检查犬粪中细粒棘球绦虫目的基因均阳性,较普通PCR法提早2d。结论 LAMP法简便、快捷、灵敏,有望成为早期诊断犬细粒棘球绦虫感染的重要手段。
Objective To determine the earliest detection date of ring-mediated isothermal amplification of canine Echinococcus granulosus (LAMP). Methods Eight domestic dogs were orally fed with protoscoleces of Echinococcus granulosus to establish the animal model of Echinococcus granulosus infection. DOG was collected 40 days after infection and LAMP was used to detect the target gene of Echinococcus granulosus in feces and compared with the traditional PCR. Results Eighteen dogs were infected with Echinococcus granulosus, and the positive gene of Echinococcus granulosus was detected by the LAMP method on the 18th day after infection, which was 2 days earlier than that of the normal PCR. Conclusion The LAMP method is simple, rapid and sensitive, which is expected to be an important method for early diagnosis of Echinococcus granulosus infection in dogs.
