目的 探讨用荧光定量聚合酶链反应方法检测脑星形细胞瘤中多药耐药 (multidrugresistance ,MDR1 )基因的表达。方法 在逆转录聚合酶链反应的基础上 ,采用荧光定量方法对经病理证实的 38例星形细胞瘤进行实时检测MDR1基因 ,并进行统计学分析。结果 复发病人MDR1基因阳性率为 88 9% ,比初发病人 (48 3 % )高 ,Ⅰ级星形细胞瘤病人MDR1基因阳性率为 2 2 2 % ,Ⅱ级病人为 66 7% ,Ⅲ级病人为 71 4%。结论 星形细胞瘤中MDR1基因的阳性率较高 ,且有原发耐药现象。荧光定量实时检测MDR1基因的方法具有较高的准确度和灵敏度 ,假阳性率低
Objective To investigate the expression of multidrug resistance (MDR1) gene in brain astrocytomas by fluorescence quantitative polymerase chain reaction. Methods Based on reverse transcription polymerase chain reaction (RT - PCR), real - time RT - PCR was used to detect 38 cases of astrocytoma confirmed by pathology. The MDR1 gene was detected by real - time fluorescence quantitative method and analyzed statistically. Results The positive rate of MDR1 gene in relapsed patients was 88.9%, higher than that in primary patients (48.3%). The positive rate of MDR1 gene in grade Ⅰ astrocytoma was 22.2%, in grade Ⅱ patients was 66.7%, in grade Ⅲ The patient was 71 4%. Conclusion The positive rate of MDR1 gene in astrocytomas is high, and there is primary drug resistance.
