以京科968、郑单958两套三联体样品为材料,分别采用CTAB和改良CTAB法提取叶片和杂交种种皮的DNA,利用maize SNP384芯片(包括384个SNP位点)进行DNA指纹分析。结果显示:玉米叶片和种皮提取DNA获得基因型数据在成功率、数据质量上一致,均可获得较好的基因型数据;通过将杂交种种皮DNA与母本叶片DNA基因型数据的比较分析,三联体样品亲子关系分析,显示种皮DNA基因型能够代表杂交种母本DNA指纹。因此玉米种皮提取DNA结合SNP芯片检测技术能够在品种权保护和品种鉴定中发挥一定作用。
Two sets of triplet samples of Jingke 968 and Zhengdan 958 were used as materials. The DNA of leaves and hybrids were extracted by CTAB and CTAB, respectively. The DNA fingerprinting of maize SNP384 (including 384 SNPs) was performed. The results showed that the genotype data of DNA extracted from leaves and seed coat of maize could obtain better genotype data with the same success rate and the same quality of data. By comparing the DNA genotypes of hybrid seed coat DNA and maternal leaves , Triad sample parent-child relationship analysis showed that the seed coat DNA genotype can represent hybrid maternal DNA fingerprinting. Therefore, the corn seed coat DNA extraction combined with SNP chip detection technology can play a role in variety protection and species identification.
