本研究以小麦山融3号为材料,克隆出基因Ta NF-YB6,其全长为435 bp,预测该基因编码蛋白分子量为16.06 k D,等电点为8.48。生物信息学分析显示,该基因与其他植物中NF-YB家族成员具有较高同源性,其中与一粒小麦Tm NF-YB5同源性最高,为74.15%。亚细胞定位结果显示,该基因编码蛋白在细胞膜和细胞核中均有分布。利用Real-time PCR对该基因在多种非生物胁迫下进行表达分析,结果表明:该基因不受Na Cl的诱导,但受PEG6000、Me JA、SA和ABA的诱导,并表现出不同的表达模式,可能在小麦抵御多种非生物胁迫过程中发挥作用。下游胁迫响应基因表达量检测显示,该基因可能在ABA依赖和ABA不依赖两条非生物胁迫响应途径中发挥作用,尤其在与RD20相关途径中发挥重要作用。
In this study, the wheat Taoyan 3 was cloned as a gene Ta NF-YB6 with a full-length of 435 bp. The molecular weight of this gene was predicted to be 16.06 kD and its isoelectric point was 8.48. Bioinformatics analysis showed that this gene shares high homology with members of NF-YB family in other plants, and has the highest homology with a wheat Tm NF-YB5 of 74.15%. Subcellular localization results show that the gene encoding protein in the cell membrane and nucleus are distributed. The results of real-time PCR analysis showed that this gene was not induced by NaCl, but was induced by PEG6000, Me JA, SA and ABA and showed different expression Mode, may play a role in the defense of wheat against multiple abiotic stresses. The results of downstream stress response gene expression showed that this gene may play a role in ABA-dependent and ABA-independent abiotic stress response pathways, especially in the pathways involved in RD20.
