目的通过研究系统性红斑狼疮(SLE)患者中白介素-27(IL-27)和骨桥蛋白(OPN)对Th17细胞分泌白介素-17(IL-17)的影响,探讨IL-27/OPN调控轴在SLE Th17细胞异常中的重要作用。方法取50例SLE患者和40例健康对照组血清,用酶联免疫吸附试验法(ELISA)检测各组血清中IL-27,OPN和IL-17的表达水平,并分析各细胞因子的组间差异以及它们间的相互关系。结果 1活动期SLE患者的IL-27,OPN和IL-17表达显著高于非活动期SLE患者和健康对照组,差异有统计学意义(P均<0.01);非活动期SLE患者和健康对照组间IL-27,OPN和IL-17水平的差异无统计学意义(P均>0.05);2血清中IL-27与IL-17水平、OPN与IL-17水平之间呈明显正相关(P均<0.01);3SLE患者的疾病活动程度评分与血清中OPN,IL-17呈明显正相关(P均<0.01)。SLE患者的疾病活动程度评分与血清中IL-27无明显相关(P>0.05)。结论 IL-27/OPN调控轴在SLE患者Th17细胞异常中起着重要的作用。
Objective To investigate the effects of interleukin-27 (IL-27) and osteopontin (OPN) on the secretion of interleukin-17 (IL-17) by Th17 cells in patients with systemic lupus erythematosus (SLE) Important role in SLE Th17 cell abnormalities. Methods Fifty sera from patients with SLE and 40 healthy controls were used to detect the serum levels of IL-27, OPN and IL-17 in each group by enzyme-linked immunosorbent assay (ELISA) Differences and their interrelationships. Results 1 The expression of IL-27, OPN and IL-17 in active SLE patients was significantly higher than that in inactive SLE patients and healthy controls (P <0.01). In inactive SLE patients and healthy controls There was no significant difference in the levels of IL-27, OPN and IL-17 between the two groups (P> 0.05) .2 Serum IL-27 and IL-17 levels, OPN and IL-17 levels were positively correlated P <0.01). The scores of disease activity in 3SLE patients were positively correlated with serum OPN and IL-17 (all P <0.01). SLE patients with disease activity score and serum IL-27 no significant correlation (P> 0.05). Conclusion IL-27 / OPN regulatory axis plays an important role in the abnormal Th17 cells in SLE patients.
