目的检测六株结肠癌细胞中Dickkopf-1(DKK1)的表达并探讨其意义。方法在6株结肠癌细胞(SW480、SW620、HT29、LS174T、LOVO、HCT116)分别应用qPCR方法检测DKK1的mRNA表达水平。用Western blot方法和细胞免疫组化方法检测DKK1的蛋白表达情况。结果荧光定量分析显示,DKK1在HT29、HCT116中的表达高于其他四株细胞(P<0.05);Western blot显示,DKK1在HCT116、HT29中的表达同样高于其他四株细胞;细胞免疫组化染色显示,DKK1在六株结肠癌细胞株中呈阳性表达。结论 DKK1在6株人结肠癌细胞系中的表达具有差异性。
Objective To detect the expression of Dickkopf-1 (DKK1) in six colon cancer cells and explore its significance. Methods The expression of DKK1 mRNA was detected by qPCR in 6 colon cancer cells (SW480, SW620, HT29, LS174T, LOVO, HCT116). The protein expression of DKK1 was detected by Western blot and immunohistochemistry. Results Fluorescent quantitative analysis showed that the expression of DKK1 in HT29 and HCT116 was higher than that in the other four cells (P<0.05). Western blot showed that the expression of DKK1 in HCT116 and HT29 was also higher than the other four cells; immunohistochemistry The staining showed that DKK1 was positively expressed in the six colon cancer cell lines. Conclusion The expression of DKK1 in 6 human colon cancer cell lines is different.
