目的:观察解毒祛瘀滋阴方对糖皮质激素造成的免疫抑制的RAW264.7巨噬细胞吞噬功能的影响。方法:通过构建YFP-PGEX-4T-1载体并转化,制备能够产生荧光的大肠杆菌BL21。应用不同浓度的地塞米松以及解毒祛瘀滋阴方干预后,加入大肠杆菌,观察巨噬细胞吞噬细菌的情况。结果:转化YFP-PGEX-4T-1载体后,大肠杆菌能够发出荧光。经不同浓度的地塞米松作用后,RAW264.7细胞吞噬能力减弱,加用解毒祛瘀滋阴方后,吞噬能力有一定程度的恢复。结论:解毒祛瘀滋阴方可以增强糖皮质激素造成的免疫抑制的RAW264.7巨噬细胞吞噬功能,这可能是解毒祛瘀滋阴方可以减少SLE患者继发感染的原因之一。
Objective: To observe the effect of Jiedu Quyu Ziyin Recipe on phagocytosis of immunosuppressed RAW264.7 macrophages induced by glucocorticoids. Methods: Escherichia coli BL21 capable of producing fluorescence was prepared by constructing a YFP-PGEX-4T-1 vector and transforming it. Application of different concentrations of dexamethasone and detoxification Quyu Ziyin Fang intervention after adding Escherichia coli to observe macrophage phagocytosis. Results: E. coli was able to fluoresce when transformed with YFP-PGEX-4T-1 vector. After different concentrations of dexamethasone, the phagocytosis of RAW264.7 cells was weakened, and the phagocytic capacity was restored to a certain extent with the addition of detoxification and stasis suppository. Conclusion: Jiedu Quyu Ziyin Prescription can enhance the phagocytosis of immunosuppressed RAW264.7 macrophages induced by glucocorticoids, which may be one of the reasons that Jiedu Quyu Ziyin Prescription can reduce the secondary infection of SLE patients.
