目的观察高氧暴露早产儿血单个核细胞血红素加氧酶-1(HO-1)蛋白表达水平及活性变化。方法选择2008年2月至2009年8月本科住院吸入高浓度氧的40例早产儿(高氧组)和吸入常压空气的40例早产儿(对照组),用Western Blot法检测血中单个核细胞HO-1蛋白表达水平、血清胆红素水平和血浆一氧化碳血红蛋白(COHb)百分比,并进行比较分析。结果与对照组相比,高氧组HO-1蛋白表达明显增加[(1.78±0.25)比(1.12±0.31),P<0.01],胆红素生成量(nmol.mg-1.h-1)及血浆COHb含量(μmol/L)均显著增加[(76.25±11.33)比(53.47±8.73),(6.73±0.42)比(2.47±0.34),P均<0.01]。结论高氧暴露可上调早产儿血单个核细胞HO-1的表达及活性。
Objective To observe the changes of heme oxygenase-1 (HO-1) protein expression and activity in blood mononuclear cells of premature infants exposed to hyperoxia. Methods Forty premature infants (hyperoxia group) and 40 preterm infants (control group) were inhaled with high concentration oxygen from February 2008 to August 2009 in our hospital. The expression of HO-1 protein, the level of serum bilirubin and the percentage of plasma hemoglobin (COHb) in nucleus were compared and analyzed. Results Compared with the control group, the HO-1 protein expression in hyperoxia group was significantly higher than that in the control group [(1.78 ± 0.25) vs (1.12 ± 0.31, P <0.01], bilirubin production (nmol.mg-1.h- ) And plasma COHb content (μmol / L) were significantly higher than those in control group (76.25 ± 11.33 vs 53.47 ± 8.73 vs 6.73 ± 0.42 vs 2.47 ± 0.34, P <0.01 respectively). Conclusion Hyperoxia exposure can up-regulate the expression and activity of HO-1 in pre-term blood mononuclear cells.
