目的:探讨芪参益气滴丸对大鼠急性心肌缺血再灌注损伤的抗氧化保护作用。方法:选取50只SD大鼠随机分为空白组、假手术组、模型组、西药组和中药组,每组10只。采用结扎大鼠冠脉左前降支方法造成心肌缺血再灌注模型,各组生理盐水或药物灌注后10天,分别测定左心室肌组织中乳酸脱氢酶(LDH)、肌酸激酶(CK)、超氧化物歧化酶(SOD)、谷肤甘肤过氧化物酶(GSH-Px)活性以及MDA含量,并采用电子显微镜观察心肌组织超微结构的变化。结果:模型组CK、LDH和MDA含量较空白组和假手术组明显升高(P<0.05),而SOD和GSH-Px含量较空白组和假手术组明显降低(P<0.05);中药组和西药组CK、LDH和MDA含量均较模型组明显降低(P<0.05),SOD和GSH-Px含量较模型组明显升高(P<0.05),中药组与西药组比较,左心室肌组织CK、LDH和MDA含量明显减低,左心室肌组织SOD和GSH-Px活力明显升高,差异均具有统计学意义(P<0.05);假手术组和空白组相比,上述指标差异无统计学意义(P>0.05)。心肌组织超微结构显示,中药组缺血-再灌注损伤区超微结构的病理改变明显改善。结论:芪参益气滴丸对缺血-再灌注心肌损伤具有抗氧化保护作用。
Objective: To investigate the anti-oxidative protective effect of Qishen Yiqi drop pill on acute myocardial ischemia-reperfusion injury in rats. Methods: Fifty SD rats were randomly divided into blank group, sham operation group, model group, western medicine group and traditional Chinese medicine group, with 10 rats in each group. Myocardial ischemia-reperfusion model was induced by ligation of the left anterior descending coronary artery in rats. Lactate dehydrogenase (LDH), creatine kinase (CK) , Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activity and MDA content were measured. The ultrastructure of myocardium was observed with electron microscope. Results: The levels of CK, LDH and MDA in model group were significantly higher than those in blank group and sham operation group (P <0.05), while the contents of SOD and GSH-Px in model group were significantly lower than those in blank group and sham operation group (P <0.05) (P <0.05), while the contents of SOD and GSH-Px in model group were significantly higher than those in model group (P <0.05). Compared with western medicine group, the contents of CK, LDH and MDA in western medicine group and western medicine group were significantly decreased CK, LDH and MDA were significantly decreased, and the activities of SOD and GSH-Px in left ventricular muscle were significantly increased (all P <0.05). There was no significant difference between the sham group and the blank group Significance (P> 0.05). The ultrastructure of myocardium showed that the pathological changes of the ultrastructure in ischemia-reperfusion injury area of TCM group were significantly improved. Conclusion: Qishenyiqi dripping pill has anti-oxidative protective effects on myocardial ischemia-reperfusion injury.
