垂体瘤是颅内常见的肿瘤,能引起内分泌功能障碍。垂体腺瘤的发病机制及调节功能的研究都集中在以下两个方面:①垂体腺瘤癌基因激活和抑制基因功能丧失的调节机制;②细胞内信号传导机制的异常。其中,Raf/MEK/ERK信号通路已被认为是疾病病因中的主要的途径之一。Raf/MEK/ERK信号通路在进化过程中是保守的,用于控制细胞生长、分化和生存。该信号通路功能的改变与人类多种类型的肿瘤有关包括垂体腺瘤。Raf/MEK/ERK信号通路在垂体腺瘤的产生中的角色,为临床干预与治疗垂体瘤提供可能性。本文对Raf/MEK/ERK信号通路在垂体肿瘤产生的作用,评估其作为垂体腺瘤的干预与治疗目标的疗效作一综述。
Pituitary tumor is a common intracranial tumor that can cause endocrine dysfunction. Pituitary adenomas pathogenesis and regulatory functions are focused on the following two aspects: ① pituitary adenoma oncogene activation and inhibition of gene function loss mechanism; ② intracellular signaling abnormalities. Among them, the Raf / MEK / ERK signaling pathway has been considered as one of the main pathways in the disease. The Raf / MEK / ERK signaling pathway is conserved during evolution to control cell growth, differentiation and survival. Changes in this signaling pathway are associated with many human types of tumors including pituitary adenomas. Raf / MEK / ERK signaling pathway in the production of pituitary adenomas in the role of clinical intervention and treatment of pituitary tumors provide the possibility. This article reviews the role of Raf / MEK / ERK signaling pathway in pituitary tumors, and reviews the efficacy of Raf / MEK / ERK as a therapeutic and therapeutic target for pituitary adenomas.
