目的 探讨GM CSF对LAK细胞抗肿瘤作用影响 ,为临床应用GM CSF提供一些实验数据。方法 按常规分离正常人外周血成单个核细胞 ,分GM CSF组和LAK细胞组进行实验 ,观察GM CSF对LAK细胞的增殖活性和LAK细胞的细胞毒活性的影响。结果 GM CSF组细胞增殖活性较LAK组细胞增殖活性高 ,增殖第 7天时 ,GM CSF组达 7 3× 1 0 6/ml细胞 ,而LAK组只有 5 9× 1 0 6/ml细胞。GM CSF组效应细胞对肝癌细胞杀伤活性为 (1 7 4 8± 1 0 6 5 ) % ,LAK组效应细胞对肝癌细胞杀伤活性为 (32 36± 7 2 9) % ;说明GM CSF组对肝癌细胞的杀伤活性较LAK组效应细胞的杀伤活性为低 (P <0 0 0 5 )。结论 GM CSF对LAK细胞抗肿瘤功能具有抑制作用
Objective To investigate the effect of GM CSF on the anti-tumor effect of LAK cells and provide some experimental data for clinical application of GM CSF. Methods Peripheral blood mononuclear cells (PBMCs) were isolated from normal human peripheral blood mononuclear cells by GM CSF and LAK cells. The effects of GM CSF on the proliferation of LAK cells and the cytotoxicity of LAK cells were observed. Results The proliferative activity of GM CSF group was higher than that of LAK group. On day 7 of proliferation, the number of GM CSF group was 73 × 106 / ml, while that of LAK group was only 59 × 106 / ml. The cytotoxic activity of effector cells in GM CSF group was (17.48 ± 1.056)%, and that of LAK group was (32 36 ± 7 2 9)%. The cytotoxicity of GM CSF group to hepatocellular carcinoma The killing activity of cells was lower than that of LAK cells (P <0 05). Conclusion GM CSF has anti-tumor effect on LAK cells
