目的:研究大半边莲多糖提取工艺。方法:多糖含量测定采用蒽酮-硫酸法;多糖提取采用水提醇沉法,以大半边莲多糖提取得率为指标进行正交试验设计,考察对大半边莲多糖提取得率的影响因素。结果:多糖含量测定的线性回归方程为A=35.602C+0.030 2(r=0.999 4),线性范围为0.002 6~0.015 4 mg.ml-1。多糖提取的最佳工艺为加入8倍量的水,70℃提取2次,每次1h。结论:为大半边莲提供了稳定可行的多糖提取工艺,多糖含量测定方法准确可靠。
Objective: To study the extraction technology of lotus polysaccharide Methods: The anthrone-sulfuric acid method was used to determine the content of polysaccharides. The extraction of polysaccharides was carried out by water extraction and alcohol precipitation method. The extraction rate of polysaccharides was investigated by orthogonal design. The influencing factors on the yield of polysaccharides were investigated. Results: The linear regression equation of polysaccharide content was A = 35.602C + 0.030 2 (r = 0.999 4) with a linear range of 0.002 6 ~ 0.015 4 mg.ml-1. The best polysaccharides extraction process is to add 8 times the amount of water extracted at 70 ℃ twice, each time 1h. Conclusion: The polysaccharides extraction process is provided for the stability of Coptis chinensis, and the determination of polysaccharide content is accurate and reliable.
