目的:探讨玄莲消癖口服液对乳腺增生模型大鼠的影响,为其临床应用提供实验依据。方法:取90只未孕雌性大鼠分为6组:正常对照组、模型组、消核片对照组、玄莲消癖口服液高、中、低三个剂量组,每组15只动物。正常对照组隔日腋下注射生理盐水,其余各组均隔日腋下注射雌二醇,共10次,复制乳腺增生病理模型。于造模的同时给予受试药物,正常对照组给予等量生理盐水。末次给药后采血测定雌二醇(E2)和孕酮(P)含量;用打孔器取第二对乳房做病理切片,观察病理形态学变化;取第三对乳房测定其体积(容积法测量)。同时于给药前、给药后均测量所有动物胸部第一对乳房直径。结果:玄莲消癖口服液20~40g/kg可明显改善乳腺组织增生性病理形态学变化其主要表现:明显减少乳腺小叶数和腺泡数,显著减小乳腺体积、腺泡腔直径及乳房直径;同时还能显著降低雌二醇水平,明显升高孕酮水平,从而明显降低E2/P值。结论:玄莲消癖口服液对乳腺增生模型大鼠的病理形态、激素水平均有明显的调节作用。
Objective: To investigate the effect of Hyunlian Hsiao-tien Oral Liquid on hyperplasia of mammary glands in rats and provide experimental basis for its clinical application. Methods: Totally 90 pregnant female rats were divided into 6 groups: normal control group, model group, ablation control group and Xuan Lian Xiao Ci oral solution in high, medium and low dose groups of 15 animals each. The normal control group was injected with saline every other day under the armpit, and the other groups were injected with estradiol under the armpit every other day for a total of 10 times to replicate the pathological model of mammary gland hyperplasia. Results: Hyun Lian Xiao Ci oral solution 20 ~ 40g / kg can significantly improve the histological changes of breast tissue hyperplasia its main manifestations: significantly reduce the number of small breast lobes and alveoli, significantly reduce the size of the breast, acinar cavity diameter and breast Diameter; while also significantly lower estradiol levels, significantly increased progesterone levels, thereby significantly reducing the E2 / P value. Conclusion: Hyun Lian Xiao Ci Oral Liquid can obviously regulate the pathomorphology and hormone level of mammary gland hyperplasia model rats.
