目的探讨毒蕈碱(M)受体及其亚型对四磨汤(SMD)诱发胃窦纵、环行肌收缩的差异性影响。方法 40只雄性SD大鼠,随机分为5组,每组8只,分别为:单纯SMD组、阿托品(Atropine)+SMD组、加拉明(Gallamine)+SMD组、4-二苯乙酰氧基-N-甲基哌淀甲碘化物(4-DAMP)+SMD组及加拉明+4-DAMP+SMD组。制备胃窦纵、环行平滑肌条,分别采用累积量加入四磨汤,以及阿托品(10-6 mol/L)、加拉明(10-6 mol/L)、4-DAMP(0.4×10-6 mol/L)、加拉明+4-DAMP孵育肌条后再累积加入四磨汤,记录平滑肌等长收缩活动,比较纵、环行肌收缩的差异性。结果四磨汤诱发胃窦纵、环行肌收缩活动增加,但两者间无明显差异(P>0.05),非选择性M受体阻断剂阿托品及选择性M2受体阻断剂加拉明对四磨汤诱发胃窦环行肌收缩活动的抑制明显强于纵行肌(P<0.05),选择性M3受体阻断剂4-DAMP及加拉明+4-DAMP对四磨汤诱发胃窦纵行肌收缩活动的抑制作用明显强于环行肌(P<0.05)。结论 M受体及其亚型参与了四磨汤对胃窦运动的调节,胃窦环行肌收缩更多的依赖于M受体及其M2亚型的激活,胃窦纵行肌收缩更多的依赖于M3受体亚型的激活。
Objective To investigate the differential effects of muscarinic (M) receptor and its subtypes on the contraction of longitudinal and circular muscles in gastric antrum induced by Si mill soup (SMD). Methods Forty male Sprague-Dawley rats were randomly divided into 5 groups (8 rats in each group): SMD group, Atropine + SMD group, Gallamine + SMD group, 4-diphenylacetoxy N-methylpiperidinium methyl iodide (4-DAMP) + SMD group and Gallamine + 4-DAMP + SMD group. Antral smooth muscle strips of the antrum were prepared, and four milliliters of soup were added in a cumulative amount, respectively. Atropine (10-6 mol / L), Gallamine (10-6 mol / L) and 4-DAMP mol / L). Gallamin + 4-DAMP incubated the muscle strips and then added four mill soup. The isometric contractions of smooth muscle were recorded, and the differences of longitudinal and circular muscle contraction were compared. Results Sirao decoction induced antral and tractorial muscle contraction activities increased, but there was no significant difference between the two (P> 0.05), non-selective M receptor blocker atropine and selective M2 receptor blocker Galalamin The inhibitory effect of Si mill soup on the contraction of gastric antral muscle was significantly stronger than that of longitudinal muscle (P <0.05). The selective M3 receptor blockers 4-DAMP and Galalamin + The inhibitory effect of contractile activity of sinus longitudinal muscle was significantly stronger than that of circular muscle (P <0.05). Conclusions M receptor and its subtypes are involved in the regulation of gastric motility by Si mill decoction. The contraction of gastric antrum is more dependent on the activation of M receptor and its M2 subtype and the contraction of antral longitudinal muscle muscle more Dependent on the activation of the M3 receptor subtype.
