为探讨亚砷酸钠(NaAsO2)对人角质形成细胞(HaCat)活力及蛋白激酶B(PKB/Akt)磷酸化活化的影响,本研究以Alamar Blue还原法检测NaAsO2(0,5,10,25,50,100μmol/L)作用于HaCat细胞6 h后细胞的活力水平;并以Western blot法检测NaAsO2(0,25,50μmol/L)作用于HaCat细胞6 h后细胞中磷酸化蛋白激酶B(p-Akt)的蛋白表达情况。结果表明,NaAsO2作用6 h,Alamar Blue还原率随NaAsO2染毒剂量的升高而显著下降,具有显著的剂量-效应关系;NaAsO2染毒组细胞内p-Akt表达水平与对照组相比均显著提高,提示砷性肿瘤的发生可能与p-Akt蛋白表达水平增高有关。
In order to investigate the effect of sodium arsenite (NaAsO2) on the activity of human keratinocytes (HaCat) and the phosphorylation of protein kinase B (PKB / Akt), Alamar Blue reduction method was used to detect NaAsO2 (0,5,10,25 , 50 and 100μmol / L) for 6 h, respectively. The expression of phosphorylated protein kinase B (p <0.05) in HaCat cells induced by NaAsO2 (0,25 and 50 μmol / L) -Akt) protein expression. The results showed that after NaAsO2 treatment for 6 h, the reduction rate of Alamar Blue significantly decreased with the increase of NaAsO2 dosage, and had a significant dose-effect relationship. The expression level of p-Akt in NaAsO2-treated group was significantly higher than that in control group Improve, suggesting that the occurrence of arsenic tumors may be related to the increased expression of p-Akt protein.
