目的:通过研究不同人群中高血压相关基因1(hypertensionrelatedgene1,HRG-1)基因多态性,以揭示高血压相关基因多态性与高血压的关系。方法:安贞医院高血压3门诊2002-06/2002-12体检的健康人1200例,纳入标准:无明确相关疾病。选符合上述标准健康人74例为对照组;同期安贞医院门诊原发性高血压患者1800例,纳入标准:符合WHO/ISH制定的高血压诊断标准。排除标准:排除糖尿病及其他心血管系统疾病、肾脏血管疾病、肾功能衰竭等疾病,并能排除盐敏感性高血压及一些单基因遗传疾病等。符合上述标准原发性高血压患者51例(高血压组)。取外周静脉血并提取DNA,设计相应的引物,PCR扩增法获得相应的产物,应用DNA测序法直接得到PCR产物的核苷酸序列后比较不同人群的相同片段的碱基组成及其特征。结果:高血压组肌酐犤(106.08±17.70)μmol/L犦和尿素氮犤(5.61±1.71)mmol/L犦明显高于对照组犤(53.04±20.03)μmol/L,(3.21±1.27)mmol/L犦(F=17.25,P=0.000;F=5.99,P=0.006),血糖、血脂等指标差异无显著性意义(P>0.05)。高血压组的收缩压和舒张压均明显高于对照组(F=17.25,5.99;P=0.000,P=0.006);高血压组的血浆血管紧张素Ⅱ(AngiotensionⅡ,AngⅡ),心钠素,内皮素及降钙素基因相关肽(calciumgenerelatedpeptide,CGRP)水平均?
Objective: To investigate the relationship between hypertension related gene polymorphism and hypertension by studying the genetic polymorphisms of hypertension related gene 1 (HRG-1) in different populations. Methods: Anzhen Hospital hypertension outpatient 2002-06 / 2002-12 physical examination of 1200 healthy subjects, inclusion criteria: no clear related diseases. In the same period, Anzhen Hospital outpatient essential hypertension patients in 1800 cases, inclusion criteria: in line with WHO / ISH developed diagnostic criteria for hypertension. Exclusion criteria: exclude diabetes and other cardiovascular diseases, renal vascular disease, renal failure and other diseases, and can rule out salt-sensitive hypertension and some single-gene genetic diseases.
