鲨鱼软骨粉(SCP)伍用“425”制剂(当归根提取物),使成鲨鱼软骨粉合剂,用以研究感染日本血吸虫3wk的BALB/c小鼠经口服用SCP合剂后4wk(相当于感染后7wk)诱导的抗血吸虫性纤维化效应及其机制。测定实验小鼠肝内血吸虫卵肉芽肿(SEG)大小和纤维化程度;用ELISA检测小鼠血清抗体IgG及其亚类IgG1水平(OD均值);应用流式细胞仪测定小鼠脾脏T细胞亚群的细胞均数和比值。实验结果:服用SCP合剂(Ⅰ组)和单用SCP(Ⅲ组)小鼠的SEG大小均显著小于感染对照鼠(Ⅳ组),且前两者肝内血吸虫性纤维化均已消失,而后者显示严重的肝纤维化。单用“425”制剂(Ⅱ组)与Ⅰ组和Ⅲ组的血吸虫卵肉芽肿减小率之间无显著性差异,但Ⅱ组仍呈现轻度的肝血吸虫性纤维化。Ⅰ组和Ⅲ组的CD4+/CD8+比值(1.70~1.88)明显高于Ⅱ组(0.95);Ⅰ组和Ⅱ组的IgG和IgG1水平均显著高于Ⅲ组和Ⅳ组。结果表明,服用SCP合剂和单用SCP小鼠,均能诱导明显的抗肝血吸虫性纤维化效应,若单用“425”制剂,则仅显示轻度的抗肝纤维化作用,其机制与细胞免疫密切相关。

The CD4 + / CD8 + ratio (1.70-1.88) in group Ⅰ and group Ⅲ was significantly higher than that in group Ⅱ (0.95). The levels of IgG and IgG1 in group Ⅰ and group Ⅱ were significantly higher than those in group Ⅲ and group Ⅳ. The results showed that both SCP and SCP mice were able to induce obvious anti-hepatic schistosome fibrosis effects. If only “425” formulation was used, only mild anti-hepatic fibrosis was shown.
