目的:探讨补精气化瘀毒方对小鼠移植性肝癌HepS的抑制作用及其机理。方法:采用小鼠移植性肝癌HepS实验动物模型,设对照组、5-氟尿嘧啶(5-Fu)组、补精气化瘀毒方高中低3个剂量组,各组连续给药7d,于末次给药24h后处死小鼠,称体重,摘瘤称重,计算抑瘤率,并对小鼠脾指数、胸腺指数、癌细胞bcl-2/bax表达、癌病理进行观察。结果:补精气化瘀毒方在连续7d120mg/kg、60mg/kg、30mg/kg的剂量下对小鼠移植性肝癌HepS的抑制率可分别达68.38%、58.08%、46.32%;补精气化瘀毒方可提高荷瘤小鼠的脾指数、胸腺指数,可使荷瘤小鼠肝癌组织中bcl-2表达水平降低,bax表达水平升高。结论:补精气化瘀毒方对小鼠移植性肝癌HepS具有显著的抑制作用,其作用机理可能与其调控bcl-2/bax基因表达,诱导癌细胞凋亡有关。
Objective: To investigate the inhibitory effect and mechanism of Bu Jing Qi Hua Du Du Fang on mouse transplanted liver cancer HepS. METHODS: A mouse model of transplantable hepatocellular carcinoma HepS was used. The control group, 5-fluorouracil (5-Fu) group, and Bu Jing Hua Hua Yu Du Fang high-medium low-dose group were given. Each group was continuously administered for 7 days. 24 hours after administration, mice were sacrificed, body weight was weighed, tumors were weighed, tumor inhibition rate was calculated, and mouse spleen index, thymus index, bcl-2/bax expression, and cancer pathology were observed. RESULTS: The inhibitory rate of Bu Jing Hua Hu Yu Du Fang at the doses of 7d120mg/kg, 60mg/kg, and 30mg/kg for 6 months was 68.38%, 58.08%, and 46.32%, respectively. Huatan Duoxue can increase spleen index and thymus index of tumor-bearing mice, and can reduce the expression of bcl-2 and the expression of bax in tumor-bearing mice. Conclusion: Bujinghuhuayudu Decoction has a significant inhibitory effect on transplanted liver cancer HepS in mice, and its mechanism may be related to its regulation of bcl-2/bax gene expression and induction of cancer cell apoptosis.
