目的观察黄芪注射液对视神经牵拉伤大鼠视网膜组织神经生长因子(NGF)及酪氨酸激酶受体A(Trk A)蛋白表达的影响。方法将48只Wistar雄性大鼠随机分为假手术组、模型组及黄芪治疗组,每组16只大鼠。横向定量牵拉法制作大鼠视神经损伤模型,假手术组仅暴露视神经,不予牵拉;术后黄芪治疗组给予黄芪注射液腹腔注射,模型组给予生理盐水腹腔注射;治疗14 d后取大鼠视网膜组织,采用蛋白免疫印迹法(Western blot)检测NGF和Trk A蛋白表达变化。结果与假手术组比较,视神经牵拉伤模型大鼠视网膜组织NGF、Trk A蛋白表达显著下降(P<0.05);给药14 d后,与模型组比较,黄芪治疗组大鼠视网膜组织NGF及Trk A蛋白表达明显增加(P<0.05)。结论黄芪注射液能够上调大鼠视网膜组织NGF、Trk A的蛋白表达,具有视神经保护作用。
Objective To observe the effect of Astragalus injection on the expression of nerve growth factor (NGF) and tyrosine kinase A (Trk A) protein in the retina of rats with optic nerve injury. Methods 48 Wistar male rats were randomly divided into sham operation group, model group and Astragalus treatment group, each group of 16 rats. The rats in the sham operation group were exposed only to the optic nerve and were not pulled. The Astragalus injection group was given intraperitoneal injection of Astragalus injection and the model group was given intraperitoneal injection of saline. After 14 days of treatment, The retinal tissue of rats was used to detect the expression of NGF and Trk A protein by Western blot. Results Compared with the sham-operation group, the expression of NGF and Trk A protein in the retina of rats with optic nerve traction injury decreased significantly (P <0.05). Compared with the model group, the retinal NGF Trk A protein expression was significantly increased (P <0.05). Conclusion Astragalus injection can up-regulate the protein expression of NGF and Trk A in the retina of rats, which has the function of optic nerve protection.
