目的探讨重组人生长激素(rhGH)对THP-1单核细胞TNF-α和IL-6分泌的影响,并研究其与NF-κB活性的相关性。方法应用ELISA检测rhGH诱导THP-1细胞培养上清中TNF-α和IL-6的分泌情况,并观察LPS和NF-κB特异性抑制剂BAY11-7082对其分泌的影响;应用荧光素酶报告基因和电泳迁移率实验(EMSA)检测NF-κB在rhGH诱导的单核细胞中的活化程度。结果 rhGH单独可以促进THP-1细胞分泌TNF-α和IL-6,抑制LPS诱导的细胞因子的分泌;BAY11-7082能显著抑制rhGH和LPS诱导的TNF-α和IL-6的分泌,NF-κB活性与TNF-α和IL-6的分泌呈现明显的相关性。结论 rhGH可通过NF-κB信号通路双向调节THP-1单核细胞TNF-α和IL-6的分泌。
Objective To investigate the effect of recombinant human growth hormone (rhGH) on the secretion of TNF-α and IL-6 in THP-1 monocytes, and to study its relationship with the activity of NF-κB. Methods The secretion of TNF-α and IL-6 in supernatant of THP-1 cells induced by rhGH was detected by ELISA. The effects of LPS and BAY11-7082, a specific inhibitor of NF-κB, on the secretion of TNF-α and IL-6 were observed. Gene and electrophoretic mobility shift assay (EMSA) was used to detect the activation of NF-κB in rhGH-induced monocytes. Results rhGH alone could promote the secretion of TNF-α and IL-6 by THP-1 cells and inhibit the secretion of cytokines induced by LPS. BAY11-7082 could significantly inhibit the secretion of TNF-α and IL-6 induced by rhGH and LPS, κB activity and TNF-α and IL-6 secretion showed a significant correlation. Conclusion rhGH can regulate the secretion of TNF-α and IL-6 in THP-1 monocytes through NF-κB signaling pathway.
