本文介绍了用生物发光技术对大鼠心肌线粒体及内质网(SR)ATP的测定.并以此来表示Ca~(2+)-ATP酶的活性.线粒体和SR样品发光强度的变异系数均在10%以内.另外,煮沸90s与不煮沸(20min内测定)灭活的酶活性基本一致.用本法测得大鼠心肌匀浆、线粒体和SR酶活性均高于传统的定磷法一倍左右.
This article describes the determination of mitochondria and endoplasmic reticulum (SR) ATP in rat myocardium by using bioluminescence technique, and expresses the activity of Ca ~ (2 +) - ATPase.The coefficients of variation of luminescence intensity of mitochondria and SR Less than 10% .In addition, the boiling 90s and not boiling (determined within 20min) inactivated enzyme activity is basically consistent with this method measured in rat myocardial homogenate, mitochondria and SR enzyme activity were higher than the traditional P Times or so.
